UCT Xtrackt® SPE Columns Cited in Horse Steroid Analysis Article

Androgenic anabolic steroids (AAs) are synthetic substances based on endogenous testosterone that can enhance muscle growth and improve athletic performance. They have quickly evolved into one of the common misused drug classes in both human and animal sanctioned sports since the first incident of athletes using AAs occurring in the mid-1950’s. The International Olympic Committee (IOC now World Anti-Doping Agency (WADA)) has banned AAs since 1976, and the use of AAs has been also prohibited in the sport of horseracing by the International Federation of Horseracing Authorities (IFHA).

The samples analyzed in the above study were hydrolyzed using β-glucuronidase/arylsulfatase and then extracted employing UCT’s Xtrackt® DAU solid phase extraction sorbent. Following this, the sample was automatically loaded on the TurboFlow Cyclone system, followed by separation on a fused core C18 HPLC column before tandem mass spectrometric detection. All analytes were rapidly detected within 10 minutes with high sensitivity (picogram to nanogram per milliliter level), and limited competing matrix interferences. The linearity range was from 0.1-10 ng/mL for nine steroids and 1.0-50 ng/mL for the others, with correlation of coefficient values over 0.995. Precision and accuracy ranged from 0.1 to 14.5% and 1.7 to 12.4%, respectively. The developed method was successfully applied to the analysis of anabolic steroids in horse urine after administration of a model drug. This study shows why analysts working in the field of animal performance toxicology turn first to UCT and Xtrackt® SPE for the very best results.
